Versatile engineering of ligand spacing on nano- and micro-assembly to regulate stem cell differentiation

Submolecular spacing of ligands regulates stem cell differentiation. In this session, we will demonstrate versatile engineering of ligand spacing on bioactive materials to control stem cell differentiation. First, we will present liganded nanoassembly with tunable local ligand spacing on Fe3O4 while keeping macroscale ligand density constant. Decreasing ligand spacing as well as falling liganded nanoassembly promotes stem cell adhesion-mediated osteogenic differentiation. Next, we will show photoreversible supramolecular liganded self-assembly of cationic azobenzene and liganded polyanions via cation-π and electrostatic interactions. Depending on irradiated light wavelength, the self-assembly can deflate or inflate, which reduces or increases ligand spacing, respectively, to regulate stem cell differentiation. Further understanding of ligand spacing can offer insight to elucidate molecular level interactions between receptors and ligands.
