A Novel Self-Degradable Prodrug Blend Thermogel System for Injectable Drug Delivery to Inner Ear

Injectable hydrogel formulations have received a lot of attention as more efficient drug delivery for inner ear treatment strategy. In this study, a novel self-degradable prodrug blend thermogel system was developed to prepare an injectable dexamethasone formulation for effective treatment of inner ear diseases. The prodrug blend thermogels were prepared by physical mixing of hexanoyl glycol chitosan (HGC) and dexamethasone conjugated glycol chitosan (GC-DEX) as a prodrug component and their physicochemical and biological properties were evaluated. They showed promising characteristics for injectable drug delivery, such as thermogelation, injectability, self-degradation, and controlled release. This formulation demonstrated enhanced drug absorption and prolonged drug concentration in the inner ear, resulting in accelerated recovery from hearing loss. Consequently, our prodrug blend thermogel system could be useful as an effective injectable formulation for inner ear drug delivery.
