Fabrication of Polydiacetylene-Silver Nanoparticle Hybrids for Fluorescence Response Augmentation in Biosensors

Polydiacetylene (PDA) is a Raman-active Pi-conjugated polymer with extensive sensing applications owing to its fluorescent (FL) feature and blue-to-red color transition in response to external stimuli. However, evolution of PDA-based biosensors remains challenging since high performance biosensors demand a substantial increase in FL signaling and a vastly improved sensitivity. Configurational changes in PDA Pi-orbital electrons with the help of optical features such as localized surface plasmonic (LSP), which is a resonant oscillation of free or delocalized electrons in nanomaterials, may lead to significantly boosted spectroscopic signals including absorption, fluorescence, and Raman effects. Hereupon, we fabricate a PDA polymer hybridizes with silver nanoparticles (Ag/PDA) biosensor using a straightforward technique. As a consequence of the resonance between the surface plasmon frequency of Ag NPs and the exciton frequency of PDA in the hybrid nanostructure, a remarkably elevated FL and Raman signal can be detected.
