Stimuli-responsive polymer nanocarriers mediated drug delivery is one of the most promising platforms for targeting antitumor treatment. In this work, by using a combination of reversible addition fragmentation chain transfer polymerization (RAFT) and ring opening polymerization, we present an efficient approach for preparing a series of disulfide-containing amphiphilic polypeptide hybrids, poly(N-isopropylacrylamide)-block-poly(3,4-dihydroxy-L-phenylalanine) (PNIPAM-b-PDOPA). After the aminolysis of the RAFT end group and the deprotection of acetyl group, the resulting hybrids were able to self-assemble into stabilized and uniform micelles in which doxorubicin and IR780 are efficiently encapsulated. The in vitro cell viability assays results illustrated that the PNIPAM-b-PDOPA hybrid bioconjugates might serve as highly biocompatible and multifunctional nanocarriers for practical applications.
