Reactive oxygen species (ROS)-responsive drug carrier have been attracted for cancer-selective drug delivery since ROS is one of the representative features of cancer cells. Thioether-bearing polymers (TEP) were synthesized to achieve ROS-responsive drug delivery. PL-loaded TEP nanoparticles (PL-TEP NPs) were fabricated to deliver hydrophobic piperlongumine (PL) into cancer cells via a nanoemulsion method. PL is one of the pro-oxidant agents that induce oxidative stress in cancer cells. PL-TEP NPs showed ROS-sensitive disassembly, which leads to efficient cytoplasmic release of PL in MCF-7 human breast cancer cells. Moreover, cancer-specific cytotoxicity of PL-TEP NPs was achieved due to the increase of intracellular ROS in cancer cells. This study demonstrates that PL-TEP NPs are promising stimuli-responsive drug carriers for intracellular delivery of hydrophobic PL.
