In vitro Osteogenic differentiation and Anti-microbial Efficacy of Dual Growth Factor/Antibiotic-loaded PCL/HA Microsphere-Hydrogel System

The needs for microspheres made of biodegradable polymers have gradually increased with the developments of bioactive delivery systems, tissue engineering, and regenerative medicine. In this study, we prepared a BMP-2-loaded polycaprolactone (PCL)/hyaluronic acid (HA) hybrid microspheres and vancomycin-loaded alginate hydrogel carrier system to be used as an injectable bone filler. This microsphere-hydrogel system showed the sustained release of both BMP-2 and vancomycin for more than one month. We evaluated the osteogenic differentiation potential and anti-microbial efficacy of this microsphere-hydrogel system. From the results, both BMP-2- and vancomycin-loaded group showed better osteogenic differentiation than the control group (w/o BMP-2 and vancomycin) and the group with single BMP-2 or vancomycin. The groups with vancomycin also showed the long-term antimicrobial efficacy.
